Actinic light effects by ECS measurements? (Leaf Photosynthesis MultispeQ V1.0)

By Ryouichi Tanaka

Hi, If I correctly understand it, the "Leaf Photosynthesis MultispeQ V1.0" protocol starts with ECS measurements. I am afraid that it might provide actinic light effects on leaf photosynthesis and affect the subsequent measurement of Phi2, PhiNPQ and PhiNO. I am speculating this because when I compared the Phi2 measurements of "Leaf Photosynthesis MultispeQ V1.0" and those... read more

Stomatal Conductance Calculation V.2 Query

By Daisy Dobrijevic

Hello all, I am new to using the MultispeQ 2.0 and I wish to calculate stomatal conductance. Is there a macro that can do this automatically or do I need to work it out from the output of the protocol? If so which outputs should I be focusing on? Using the RIDES protocol (believe this is the correct one?!), I receive the following values (along with a whole host of others... read more

OJIP transients and P deficiency - can it be measured with the MultiSpeQ?

By Anton Wasson

Hello All, I am a newcomer to MultiSpeQ, PhotosynQ, and photosynthesis research in general. I work on root development and P deficiency. I recently read two papers which suggest that leaf P content and deficiency can be predicted from a multivariate analysis of the I step of an OJIP transient: Carstensen, A., Herdean, A., Schmidt, S. B., Sharma, A., Spetea, C., Pribil, M.... read more

MultispeQ beginner questions

By Robert Walters

All, I am just getting started with MultispeQ v.1 so please bear with me. Q1: My instrument has a set of SPAD recalibration cards. How, and when, should these be used? I find no guidance anywhere. Q2: It is not clear what exactly the trace parameters ECSt, vH+, gH+, are estimating, and their relationship to other parameters. Per the tutorial, these describe the accumu... read more

The rate of turnover of the cytochrome b6f complex

By Andriy Herts, Nataliia Herts

Hi. In the protocol RIDES, I see possibility measuring of "ATP synthase conductivity, proton motive force parameters through DIRK(ECS)" and "The rate of turnover of the cytochrome b6f complex through DIRKP(700)" Both based on Dark-interval relaxation kinetic (DIRK). For estimate ATP synthase we have ECSt mAU, vH+, gH+ parameters in protocol and visualization setting thro... read more

Burning brake smell

By Tshegofatso

Hi, My front Outer drivers side pad worn completely down, steel on steel, yet inside pad was half meat, and so was passenger side. I replaced both pads and rotors. Afterwards had a burning smell coming from front drivers side wheel, and it was getting very hot, to the point that the lug nuts were hot to the touch. I have now replace the entire caliper for both sides, I ble... read more

Operator and device position (facing the sun / back to the sun) during the measurement

By Miriam Fernández

I am taking measurements with Multispeq in a quite wide barley field where plants present erect leaves. I would like to know in which side of the plot should I stand to take the measurements, as one side is shaded and the other faces the sun, even if I try to measure in central hours when the sun is high. What would be more accurate? I understand the part about not movin... read more

Recommendations or experiences with any inexpensive android phones.

By Craig Whippo

More than half of my students have iphones. For these students, I purchased a couple of neutab android tablets that have built-in gps to connect to V1.0. These tablets are too big to hold in one hand, screen glare is an issue, and the blue-tooth connectivity is easily disrupted. I'm thinking about replacing these tablets with some inexpensive android phones. Does anyon... read more

PCA and data analysis

By Andriy Herts, Nataliia Herts

I try using PCA (Principal component analysis) in the analysis of data. For example, Phaseolus vulgaris + molybdenum (difer. concentration) in lab (fig.1) and field (fig.2). Interesting not only influence of molybdenum as well as changes in correlated variables in lab and field plant especially!!! Interestingly grouped options. Is not it. Who uses and how interpret PCA? ... read more

By Weizhen Liu

After updating to latest Leaf Photosynthesis MultispeQ V1.0 protocol, I cannot find qL, qP, ECSt, vH+, gH+ and many other parameters that previous Leaf Photosynthesis MultispeQ V1.0 protocol provided in data viewtable in spreadsheeteither online or download to local computer. Does anyone else have the same situation as me? How can I get these data? Thanks.

How to identify and use already developed protocols by different people?

By Habib-ur-Rehman Athar

Different people developed different protocols and and long list is available. I selected to use but none of them is working. Either they are not developed or I am not properly using. I want to use NPQ quenching analysis, DIRK protocols. I am not familiar with program coding so I want to use already developed protocols by other peoples. MultispeQ has only one functional pro... read more

How to interpret data correctly from field?

By Andriy Herts, Nataliia Herts

The parameters (Phi2, PhiNPQ, PhiNO) are dependent on the level of lighting. If you measure plant in the field for several hours, for example from 10 am to 1 pm . (Fore example: Option 1 - in 10.00 am; Option 2 - in 10.20 am .... Option 10 - in 1 pm et all. ) How to correctly interpret data for option 1 and 10? PAR in 10.00 am - 600, but in 1 pm 1600 unit . Phi2 in10 a... read more

Carbon evolution and active carbon in soil (measurement methods)

By Andriy Herts, Nataliia Herts

Hello! I want to start the study of soil. Do I understand correctly: 1. I can measure carbon evolution from soil - soil (on the field, for example)+ chamber+CO2 sensor. After this converts CO2 ppm in kg C m-2 day (don't need of calibration ) 2. I can measure active carbon - example of the soil + chamber+CO2 sensor + potassium permangenate solution (need in calibration) Th... read more

Making a balance that interfaces with the photosynq app - where to connect outputs on Multispeq board?

By Steven Vanek

Hi, I think this might be for Greg or Dan -- I have a little balance rig with a load cell and amplifier that has a nice linear output 0 to 50 g which outputs to ~0 to 4.25 volts -- I can increase the range to 300g by changing a gain resistor. Where could I hook this on the multispeq arduino board? I would need ground, 5V power, and the sensor output. I got this off an ar... read more

Problems with data visualization after measurement

By Oscar Goni

Hi! My labmate and me are observing a curious problem with the data visualization. Suddenly, we can't see numerical values after performing either a quick or a project measurement. Instead that, we can see a graph and the values NaN (see attached file). However, if we keep the project measurement, we can see the numerical values in our Dashboard. It is a strange issue beca... read more

Light leakage

By Chris Fastie

Are there data about the effect of light leaking into the chamber during measurement? If the sun is shining right on the gap, there could be variable amounts reaching the light guide. I was thinking about attaching a little black felt skirt that blocked at least the direct sunlight from hitting the gap. But maybe the light guides are not sensitive to light coming from the s... read more
