Measuring FvFm and light derived parameters

Hello, we would like to measure FvFm after dark adaptation and then light-adpated parameters such LEF, ECS, etc. I am aware that there many protocol for measuring FvFm and the other parameters. However, would you reccommend one that measures both? or, would it be possible to run two protocols in series? For example, I am thinking of using Rodrigo's "10 - Fv/Fm - v1 - OK" and Dave's "RIDES_light_potential_ECS"? Any feedback would be very welcome. Gonzalo

Hi Gonzalo, yes, yo have some options to do that. You can use both protocols in the same project to take measurements in series, or in case you want to run the protocols directly from the chrome app you can pick the first protocol, press ADD (the second botton on your right) and look for the second protocol and press add again (I attached a file with a screen print of what you'd get). Protocols are shown at the top side, and you will see a run botton at the right edge. You should use that one to run both protocols in series. Rodrigo, thanks for your reply. We tried to “Add” both your protocol “10 – Fv/Fm – v1 – OK” and the “RIDES_light_potential_ECS” using the “older” version of the Chrome PhotosynQ Add but they did not run together (Note that the newer version of the App does not have the “Add” option anymore). We also tried to run them together as part of the a “project” but it did not work either. However, both protocols worked well when run individually. I am not sure how we can find out if “compatible” protocols can be run in series at the moment. Perhaps, would it be easier to write one protocol that can do both?

Hey,
it is correct, that the new Desktop App has no longer the function to chain multiple measurement protocols together. One of the reasons is the introduction of protocol sets. I'm not 100% sure, but I think chaining a protocol like the "RIDES_light_potential_ECS" which is using a protocol set with another protocol is not working. Also we have seen that chaining multiple long protocols together can cause memory issues on the Instrument side. I would recommend modifying the "RIDES_light_potential_ECS" and include the function of the "10 – Fv/Fm – v1 – OK" protocol into it.

Hi Gonzalo, Are you trying to take a series of measurements of Fv/FM with different PAR values? I recently wrote up a tutorial on how to do these kinds of protocols using RIDES protocol_sets. If think it should be a good prototype for what you need, but please give me some more details. Dave

Sure. I made two protocols/projects as an introduction to the approach. These have quite a lot of annotation to explain how it all works.

Dave, we are running a drought experiment and we wanted to use FvFm as a "diagnostics" for plant health. In addition, I wanted to calculate the LEF. For what I could read in the description of your new protocol "https://photosynq.org/protocols/simple_fluor_light_curve_with_recovery_adv"; I assume the first reading would do it as "so that the 1st trace will be in dark," but it would also give recovery parameters, which is great.
